Oxidative Damage

Cellular responses to oxidative stress are of interest to investigators of a wide variety of human diseases such as cancer, diabetes, atherosclerosis, stroke, Alzheimer’s, many auto-immune diseases, and aging. Oxidative stress occurs when the levels of reactive oxygen species (ROS) increase dramatically in the cell leading to oxidative damage.  The estimated frequency of oxidatively-induced DNA lesions is approximately 104 lesions per cell per day. The most abundant DNA lesion generated following exposure to hydroxyl radicals is 8-oxo-dG. Biomarkers frequently used to measure cellular defensive responses against oxidative stress include superoxide dismutase (SOD), glutathione reductase, glutathione peroxidase, and glutathione (GSH).

      Superoxide Dismutases (SOD) catalyze the dismutation of the superoxide radical (O2 -) into hydrogen peroxide (H2O2) and elemental oxygen (O2). This provides an important defense against the toxicity of superoxide radicals. SOD1 and SOD2-deficient mice spontaneously develop liver cancer and are prone to develop tumors, whereas over expression of SOD protects tumor cells from apoptosis.  Trevigen offers two different Superoxide Dismutase assay kits to fit your needs.

      The HT Superoxide Dismutase 96 well Plate Assay Kit utilizes WST-1, which is converted to WST-1 formazan by xanthine oxidase (XOD).  The WST-1 formazan product is read at 450 nm in a plate reader.  The Superoxide Dismutase Cuvette Assay Kit employs NBT, which is converted to NBT-diformazan by XOD.  The NBT-diformazan product is read at 550 nm in a spectrophotometer. Both assays are free of interference by other catalytic activities, and unlike some other assay kits for SOD, these systems are not greatly disturbed by trace metals.  In addition, both kits are ideal for assaying SOD in mammalian cell or tissue lysates.  Please click the links below for more information on each assay.

        Glutathione (GSH) is an important intracellular tripeptide and a known antioxidant.  The thiol group within the cysteine residue acts as a reducing agent to assist reduction enzymes such as glutathione peroxidase to convert the highly reactive hydrogen peroxide to water.  In the process, the glutathione is oxidized to glutathione disulfide (GSSG).  The reverse reaction takes place using the glutathione reductase enzyme and NADPH as the reducing agent to reduce GSSG to GSH.  Trevigen offers several different glutathione assay kits depending on which reaction you wish to study.

        The Glutathione Reductase Assay Kit (Cuvette) measures levels of glutathione reductase in cell lysates, erythrocyte lysates, and tissue homogenates.  This is the cuvette version of the 7513-500-K.

        The HT Glutathione Assay Kit (Total Glutathione) quantifies the amount of glutathione in mammalian cells, tissue, blood, plasma and other bodily fluids using a 96 well plate.

        The HT Glutathione Peroxidase Assay Kit measures the amount of glutathione peroxidase in plasma, erythrocyte lysates, tissue homogenates, and cell lysates using a 96 well plate.

        The HT Glutathione Reductase Assay Kit (96 Well Plate) measures levels of glutathione reductase in cell lysates, erythrocyte lysates, and tissue homogenates.  This is the 96 well plate version of the 7510-100-K.
