PARP, PARG and Tankyrase
Poly (ADP-ribose) polymerases (PARPs) are a family of related enzymes that catalyze the transfer of ADP-ribose onto target proteins. PAR synthesis and subsequent degradation by poly-(ADP-ribose) glycohydrolase (PARG) are important roles in DNA repair. The discovery that certain tumors are defective in homologous recombination repair and depend on PARP-mediated DNA repair for survival suggests PARP and PARG as potential targets in cancer therapy. In addition, Tankyrase 1 (PARP 5a) plays an important role in telomere elongation. Screening tools for potential PARP, PARG and Tankyrase inhibitors are important for drug discovery.

Once inhibitors of PARP 1 have been identified using in vitro screening assays, their in vivo activity in tissue or peripheral blood mononuclear cell extracts can be measured using Trevigen’s PARP in vivo Pharmacodynamic Assay II Kit. The PARP/Apoptosis Assay Kits measure decreasing levels of PARP 1 activity as cells move through the apoptotic pathway in cell line extracts. Tankyrase 1 (PARP 5A) Activity Assay Kits are also available for the screening of Tankyrase 1 inhibitors and determining IC50 values.

Poly (ADP-ribose) polymerases (e.g. PARP-1, PARP-2), and the SIR2 family of NAD+-dependent histone/protein deacetylases, use NAD+ as a substrate for their function. In addition to NAD+, Trevigen offers Biotinylated NAD+ and Fluorescein-NAD+ for detection of PARP activity.

PAR and PARP Antibodies (7)
The PAR monoclonal and polyclonal antibodies are specific for PAR polymers synthesized by a variety of PARP isoforms (PARP 1, 2, 3, tankyrase, vPARP, and others).
The PARP monoclonal antibody can be used for analysis of PARP protein levels and cleavage of PARP in apoptotic cells. It recognizes both the full-length protein (116 kDa) and an 85 kDa cleavage fragment associated with apoptosis.
